Now that we can setup Sub-Groups...How can I build a 2-Tier Grouping using (Groups & Sub-Groups)

Unfortunately I don't think you will be able to do this. We have been doing a lot of testing on this since it was in Preview Sandbox but the group hierarchy is not brought over to the user or project. You only have Home Group and Other Groups fields. So if you were doing a User report to see who was in what Business Unit and then What Department, you can't. :(
