When it comes to built-in or script-based profile variables in Target, Do we need to activate them first following path Target -> Administration -> Response Tokens? And, How long do they take to get into action so that I can see them populating? Is that the reason that I am unable to see category affinity under Network tab in the developer console?

Yes, To see the response tokens working in action, you need to activate them first on the path you mentioned and it will be delivered immediately only through Target responses delivering an activity.
For category affinity, based on the frequency and recency of visits to your product categories it will be delivered.

Category Affinity might not have values on the initial page load, that is why it is showing an empty value. You first need to visit some categories on your site and based on your visits, Target calculates the user’s affinity for each category.
