I made an excel file full of report builder requests two weeks ago, and if I reopen it and log in to report builder, there seem to be no more report builder requests. I do see the old results, but I cannot update the file, and if I click 'manage' in the add in tab, it says there are no requests.
Any ideas on how to solve this (other then remaking the excel)?

Try to disable and re-enable Report Builder addin and see if it works. Also check for your firewall settings, if they are conflicting with it.

When you right click on the data that you have extracted, do you see the edit request option or not?
