One of our Sales team members would rather not receive individual email alerts when a new lead is assigned to them. However, the rest of the team would like to receive the alerts. What's the best way to exclude 1 person from sending the alerts?

I'm not sure how your alerts are set up but I would think that a constraint would be the easiest way to filter out the one person who doesn't want alerts. Something like this in a flow step:
That way you can make sure alerts go to all the other lead owners but not the one who would rather not receive any.
