How do I add or remove templates so that it is enabled in Preset when generating output

To make your new template visible in the Layout dropdown under Preset > Layout:

Go to Tools > Guides > Folder Profiles > {your-folder-profile} > Templates and add/remove templates as needed.

If I am understanding your query correctly, you need to add your template into the folder profile. The link below will explain how to add your created template into the global folder profile, and then you will be able to access it globally.
